Article in Press: Recent advances in delivery strategies for CRISPR-based genome editing | #MolecularMedicine #CRISPR #GenomeEditing link.springer.com/article/10.1...

Fig. 2 Multiple strategies for CRISPR delivery. A Clinical CRISPR-based genome editing can be accomplished by either in vivo or ex vivo delivery strategies. The ex vivo strategy entails the extraction of target cells from the patient, followed by precision genome editing within a controlled biomanufacturing environment, and concludes with the reinfusion or engraftment of the modified cells. Conversely, the in vivo strategy involves the direct administration of the CRISPR machinery
into the patient, requiring the diverse delivery methods to successfully navigate physiological barriers to reach the designated tissues. B Four primary categories of vectors utilized for the intracellular CRISPR delivery. Viral vectors leverage the highly evolved infectious pathways of viruses, such as adenovirus, adeno-associated virus, and lentivirus. Physical methods actively induce transient cellular porosity, encompassing microinjection and electroporation. Nanoparticle-based methods, provide high....

Fig. 2 Schematic representation of in vivo pancreatic organogenesis and in vitro SC-islet differentiation. A In vivo pancreatic development. Beginning from the pluripotent blastocyst stage, cells undergo gastrulation to form the definitive endoderm, which subsequently forms the primitive gut tube. Pancreatic buds emerge from the posterior foregut comprising multipotent pancreatic progenitor cells. The epithelium then expands and segregates into tip and trunk domains. Tip cells give rise to acinar progenitors, whereas ductal and endocrine progenitors arise from trunk. Endocrine progenitors differentiate into all endocrine pancreatic lineages that constitute the islets of Langerhans. B In vitro differentiation of SC-islets. Differentiation protocols recapitulate key steps of in vivo development. Through the sequential addition of signaling pathway activators and inhibitors (red), pluripotent stem cells are guided through successive progenitor stages towards functional islet-like clusters

Article in Press: Targeting extracellular caspase-1 attenuates sepsis-induced acute lung injury | #MolecularMedicine #eCASP1 #AcuteLungInjury #Sepsis #Inflammasomes
@NorthwellHealth
#FeinsteinInstitutesforMedicalResearch link.springer.com/article/10.1...

Fig. 6 Summary of findings. In sepsis, inflammasomes activation promotes the cleavage of caspase-1 and the extracellular release of its p20 subunit through GSDMD-dependent membrane processes. This release may occur in association with GSDMD pore formation as well as membrane disruption during lytic cell death (pyroptosis). Once released, eCasp-1 acts as a potent DAMP. Therapeutic intervention
with the inhibitory peptide C16, which specifically target
eCasp-1, effectively attenuates systemic inflammation, ALI,

Article in Press: Research advances on CD47 in digestive system cancers: from molecular mechanisms to clinical translation #MolecularMedicine #CD47 #Cancer #DigestiveSystem link.springer.com/article/10.1...

Fig. 1 Upstream regulation of CD47 and the CD47–SIRPα signaling axis in digestive system cancers. A Examples of CD47 regulation in digestive system cancers include Galectin-3 in gastric cancer, the LINC00460/miR-186-3p/MYC network in colorectal cancer, and miR-133a in esophageal cancer. B CD47 binding to SIRPα induces ITIM phosphorylation and recruits SHP-1/2, thereby inhibiting macrophage phagocytosis. Reduced recruitment and activation of non-muscle myosin IIA is one downstream effect. C CD47 blockade can increase macrophage-mediated tumor-cell phagocytosis. Tumor-antigen uptake and MHC class I cross-presentation by dendritic cells may also promote CD8 + T-cell priming.
