This piece is accompanied by a multi-stakeholder consensus study on practical fixes for poor antibody validation in research plos.io/3Ui0czP

Researchers often select antibodies based on previous use or peer recommendations. This study shows that poor #antibody validation contributes to substantial avoidable consumption of animal and human biological samples, proposing interventions to improve #reliability.

Self-reported antibody validation practices among researchers. Distribution of IWGAV pillar usage among survey respondents (n = 107). Of 107 respondents, 77 (72.0%) used at least one IWGAV-aligned validation strategy, while 30 (28.0%) used no IWGAV pillars, relying instead on technical controls only (n = 26) or performing no validation (n = 4). Among IWGAV users, 44 used a single pillar, 22 used two pillars, 10 used three pillars, and 1 used five pillars.

Poorly validated #antibodies misdirect research, wasting an estimated billion dollars and millions of biological samples each year. In this multi-stakeholder Delphi consensus study, experts agree on priority practical fixes, and who should deliver them, by 2030.

Better antibodies, better science.

Poorly validated antibodies generate misleading results, waste time and money.

Now, 2 articles and 1 primer in @plosbiology.org

Have you seen this WB from ThermoFisher?

Still anti-p53 antibody, but another clone (MA-12648 instead MA5-12557).

It also looks like that the band in lane 3 was rotated and reused in lane 4.

Moreover, it seems that same band was used for the 2 different clones.

TREM2+ macrophages have rapidly emerged as key regulators in diseases.

Studying them requires specific antibodies.

Among 14 tested antibodies in KO cell lines by WB, 5 are specific.

Human TREM2 antibody screening by western blot on lysate.

10 μg of protein from PMA-treated THP-1 WT and TREM2 KO were processed for western blot with the indicated TREM2 antibodies. The Ponceau stained transfers of each blot are presented to show equal loading of WT and KO and protein transfer efficiency from the acrylamide gels to the nitrocellulose membrane. Antibody dilutions were chosen according to the recommendations of the antibody supplier. Antibody dilutions used: ab209814** at 1/1000; ab318262** at 1/1000; ARP49413_P050 at 1/1000; MAB11618** at 1/200; MAB17291R** at 1/500; 29715* at 1/1000; 55739** at 1/1000; 91068** at 1/1000; GTX637386** at 1/1000; GTX637387** at 1/1000; ZRB2124** at 1/1000; 68723-1-Ig* at 1/2000; 83438-6-RR** at 1/2000; MA5-51494** at 1/200. Predicted band size: 25.4 kDa. **= recombinant antibody, *= monoclonal antibody.
