Recent longevity studies have used comparative genomic analysis to reveal gene markers with the potential to slow aging and inhibit cancer in long-lived species. Vaisvil et al. report the first genome sequence of the 194-year-old Aldabra giant tortoise Jonathan, the oldest known living land animal. DNA methylome analysis showed that comparatively lower-entropy regions in Jonathan’s methylome were linked to higher-fidelity gene transcription and more efficient DNA repair, energy production, and RNA processing. These findings support a model for aging where efficient energy production supports robust DNA repair, thus preserving genomic integrity and potentially contributing to improved longevity.

The Guliya ice cap (GIC) is the largest body of ice in the western Kunlun Mountains in the Tibetan Plateau. Ice cores drilled on the GIC preserve the record of past climate change in the region, an area which provides water for several billion people in Central and South Asia. Thompson et al. analyzed ice cores drilled in 1992 and 2015 and used cosmogenic isotopes and Asian monsoon records to construct an accurate climate timescale extending back 128,000 years, the oldest ice-core-derived climate record from the Tibetan Plateau. The timeline supports the presence of glacial-stage ice in the GIC, confirming that GIC ice is over 100,000 years old.

In developing ACE, Yang et al. integrated acoustofluidic manipulation into a conventional plunge-freezing workflow, preserving the structural integrity of samples while improving imaging quality. The sound waves eliminate contamination as no physical tools are needed, helping prevent damage when capturing images and ensuring a more uniform distribution of nanoparticles. ACE provides a method to improve specimen quality and imaging efficiency and expand structural studies of challenging biological systems.
