The primary sources of reactive oxygen species (ROS) in the cardiovascular system include mitochondrial dysfunction, NADPH oxidases (NOX), xanthine oxidase (XO), and uncoupled endothelial nitric oxide synthase (eNOS). A Triggers, such as chronic inflammation and hyperglycemia, cause mitochondrial membrane potential loss, which induces leakage of free radicals like superoxide anions and hydroxyl radicals. B In the presence of xanthine oxidase, hypoxic conditions may also activate the conversion of xanthine and hypoxanthine to uric acid, which is a potent pro-oxidant. C In addition, inflammatory mediators activates NADPH oxidases (NOX) to produce free radicals. D Under pathological conditions, such as dyslipidaemia or renal impairment, eNOS becomes uncoupled and produce superoxide instead of nitric oxide, in the presence of cofactors like as tetrahydrobiopterin (BH4).
